Specificity Statement: This antibody is specific for gangliosides GM3 (N-acetylneuraminic acid), GT1b, GD3, GD1a, and GQ1b but does not react with other glycolipids such as lactosylceramide, GM3 (N-glycolylneuraminic acid), GM2, GM1, GD2, GD1b, 9-o-Ac-GD3, SSEA-1, or sialylparagloboside.
Application Notes (Clone): The specificity of the original antibody (human IgM) was assessed using ELISA, revealing that it reacted with gangliosides in the following order: GM3 (N-acetylneuraminic acid) > GT1b > GD3 > GD1a > GQ1b. A recombinant version of the antibody was generated, and its biological activity was evaluated. CDC activity tests demonstrated efficacy against MeWo, M12, M14, and G-361 melanoma cells, while showing no significant CDC activity against human normal skin fibroblasts, human adenocarcinoma, human neuroblastoma, or Vero cells. These findings confirm that the recombinant antibody retains the biological activities of the original AKH11 antibody (EP0492409A1).
