Antigen Long Description: The original antibody was isolated by using monkeypox virus A35 protein as bait to isolate specific single memory B cells from the peripheral blood of two people born before 1980 by flow cytometry. The two monoclonal antibodies recovered were cloned and identified using PCR and nested PCR techniques.

Specificity Statement: This antibody binds the C-terminal domain of A35 protein (163-TSDYQDSDVSQEVRKYFCT-181) of the monkeypox virus and also cross reacts with A33 protein of vaccinia virus. Mpox is an infectious disease caused by the monkeypox virus which can be transmitted to humans through physical contact with someone who is infectious, with contaminated materials, or with infected animals. It can cause a painful rash or mucosal lesions which can last 2–4 weeks, enlarged lymph nodes and fever. Monkeypox mortality rates range from 0%-11% in the general population and are higher in young children. In Q3 of 2024, the WHO declared monkeypox a Public Health Emergency of International Concern. This product serves as a powerful tool for related research efforts.

Application Notes (Clone): The original human IgG antibody binds monkeypox virus A35 protein with a very high binding affinity of less than 1 pM as measured by SPR. This antibody can bind MPXV-A35 and VACV-A33 in an ELISA. The epitope characterization of this antibody was performed using flow cytometry by constructing several C-terminal truncated A35 variants and expressed them individually on the surface of 293T cells by transient transfection. This antibody can recognize monkeypox virus A35 protein and vaccinia virus A33 protein in a western blot assay. This antibody can be used to recognize live vaccinia virus (VACV) in an immunofluorescence assay. The cellular localizations of A33 in the VACV-infected Vero E6 cells was clearly visualized using this antibody (PMID: 37230974).
