Background: PAK2, also known as P21 protein (Cdc42/Rac)-activated kinase 2, belongs to the group I of the PAK family of serine/threonine kinases. PAK2 can be found both in the cytosol and nucleus of most cells. It can be activated by cellular context dependent partners, and plays a role in several crucial cellular processes, such as cell proliferation and growth. PAK2, in particular, is linked to responses to caspase-mediated apoptosis. The molecule is cleaved in response to cell stress in a caspase-dependent manner, but its non-cleaved form seems to have anti-apoptotic effects. Amongst the PAK proteins, it is the most common protein mutated in cancer, particularly common in lung cancer. Once activated it can modulate the activity of proteins like c-Jun to stimulate melanoma cell growth, or caspase-7 to inhibit apoptosis in breast cancer cells. Inhibition of PAK2 activity or expression has shown promising results in research studies and may represent a new therapeutic approach in cancer therapy.
Contraindications: The final concentration of DMSO in the assay should not exceed 1%.
