Applications: Study enzyme kinetics and screen small molecular inhibitors for drug discovery and high throughput (HTS) applications.
Background: CHK2 (checkpoint kinase 2, also known as CHEK2) is an evolutionarily conserved serine/threonine protein kinase that operates as a cell cycle checkpoint in response to DNA damage. Thus, it is rapidly phosphorylated and activated upon DNA damage. It is known to phosphorylate and inactivate phosphatase Cdc25C, thereby preventing entry into mitosis. CHK2 also stabilizes tumor suppressor TP53 and is a key player in the ATM DNA damage response and repair pathway. CHK2 mutations are observed in Li-Fraumeni syndrome, an inherited predisposition to cancer. Mutations in CHK2 are also linked to various spontaneous types of cancer.
Contraindications: The final concentration of DMSO in the assay should not exceed 1%.
Description: The CHK2 Kinase Assay Kit is designed to measure CHK2 kinase activity for screening and profiling applications using ADP-Glo® as a detection reagent.
Storage Stability: This assay kit will perform optimally for up to 6 months from date of receipt when the materials are stored as directed.
Supplied As: The assay kit comes in a convenient 96-well format, with enough purified recombinant CHK2 kinase, kinase substrate, ATP and kinase assay buffer for 100 enzyme reactions.
Uniprot: O96017
Warnings: Avoid freeze/thaw cycles.
Biosafety Level: Not applicable (BSL-1)
