Background: This is a standard is prepared by pooling serum obtained from a large number of qualified (healthy, non-medicated) adult blood donors. The determination of each of the subclasses was repeatedly done by ELISA using monoclonal antisera to human IgA subclasses and isolated polyclonal IgA1 and IgA2 from normal human serum, as primary standards. Different IgA1 and IgA2 paraproteins were also used and the results did not differ from the polyclonal standards. To estimate the inter-assay variation, the secondary standard NOR-04 and he primary standards were screened several times on various days. The serum contains assigned values for each of the subclasses of IgA and for total IgA: IgA1--2.00 mg/ml. IgA2--0.20 mg/ml.Total IgA--2.20 mg/ml. Human Standard Serum NOR-04 has been tested and found negative for HbsAg and antibodies to HIV (HTLVIII). After filtration, accurate volumes of 0.25 ml are filled into vials. Human Standard Serum NOR-04 is intended for use as a calibrator to prepare standard curves in quantitative determinations of the subclasses of human IgA.
