Applications: Screen or titrate anti-spike antibodies or inhibitors; use as antigen control in assay design and optimization.
Assay Conditions: The ELISA was performed by coating 100 µl of ACE2-Fc at 2 µg/ml in PBS at 4°C overnight. The plate was washed in PBS and blocked with 200 µl of 1% BSA in PBS for 1 hour at room temperature. Increasing amounts of Spike Trimer BF.7/BA.5.2.6/BF.11 were added to the plate (serially diluted in blocking buffer). The plate was washed in PBS before adding 100 µl of AffiniPure Peroxidase-conjugated Anti-His-Tag antibody (Jackson Immuno Research #300-035-240) diluted 1:5,000 in blocking buffer for 1 hour at room temperature. The plate was washed with PBS and 100 µl of MB/E Ultra-Sensitiv HRP Substrate (EMD Millipore #ES022-500ml) was added for 5 minutes at room temperature. The reaction was stopped with 100 µl of 0.6N H2SO4 and read at 450 nm.
Description: Recombinant SARS-CoV-2 Spike protein in its homotrimeric form, containing S1+S2 subunits. This protein corresponds to SARS-CoV-2 Omicron Variant BF.7/BA.5.2.6/BF.11 and contains the Omicron Spike mutations listed below. The construct also contains a C-terminal His-tag. The recombinant protein was affinity purified.
