Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
Assay Conditions: Enzyme reactions include 40 mM Tris pH 7.4, 20 mM MgCl2, 0.1 mg/mL BSA, 10 µM ATP, 0.2 mg/mL casein, 3 mM DTT, and various amounts of ALK2 incubated 45 minutes at 30 °C. Followed by addition of 25 µL of BPS ADP-Glo reagent incubated 45 minutes at room temperature. Followed by addition of 50 µl of BPS Kinase Detection Reagent incubated 45 minutes at room temperature. Luminescence was measured.
