Specificity Statement: This antibody binds human CD30, also called TNFRSF8, is a member of the tumor necrosis factor receptor superfamily. It may play a role in the regulation of cellular growth and transformation of activated lymphoblasts. It is characteristically expressed in certain hematopoietic malignancies, including anaplastic large cell lymphoma and Hodgkin lymphoma, among others.

Application Notes (Clone): This antibody was used to study the expression of CD30 and other cell surface markers on YT lymphoma cells using flow cytometry. The binding of this antibody to CD30 induces inhibition of cytotoxicity of YT cells for Raji cells. CD30 stimulation by AC10 down-regulates CD28 expression on YT by >80%. C10 treatment of YT cells additionally downregulates the expression CD45 and up-regulates IL-2R p55 (PMID: 8245437). This antibody was used for the generation of an antibody-drug conjugate called Adcetris (Brentuximab vedotin). The chimeric version of the antibody cAC10 was conjugated to the cytotoxic agent monomethyl auristatin E (MMAE) via a valine-citrulline peptide linker to create the antibody-drug conjugate cAC10-val-cit-MMAE. The release of cytotoxic tubulin modifier MMAE into the cytosol induced G2/M-phase growth arrest and cell death through the induction of apoptosis. This ADC was approved by the FDA for the treatment of Hodgkin lymphoma and systemic anaplastic large cell lymphoma (PMID: 12714494).
