Applications: Useful for the study of bromodomain binding assays, screening inhibitors, and selectivity profiling.
Assay Conditions: BRD3 protein is incubated with biotinylated acetyl-histone peptide substrate (Cat. #33000) in BRD assay buffer (Cat. #33001) in a 10 µl reaction for 30 min at RT. AlphaLISA® GSH acceptor beads (PerkinElmer) are added, followed by Alphascreen® Streptavidin- conjugated donor beads (PerkinElmer). After 1 hour incubation at RT, Alphacounts are measured.
Description: Human Bromodomain containing 3, also known as BRD3 or RING3L, GenBank Accession No. NM_007371, a.a. 29 - 417 corresponding to bromodomains 1 and 2, with N-terminal GST-tag, MW = 70 kDa, expressed in an E. coli expression system.
Format: Aqueous buffer solution
Formulation: 40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, and 20% glycerol.
