Applications: Useful for the study of bromodomain binding assays, screening inhibitors, and selectivity profiling.
Assay Conditions: BRD4 protein is incubated with biotinylated acetyl-histone peptide substrate (Cat. #33000) in BRD assay buffer (Cat. #33001) in a 10 µl reaction for 1 hour at RT. AlphaLISA® GSH acceptor beads (PerkinElmer) are added, followed by Alphascreen® Streptavidin -conjugated donor beads (PerkinElmer). After 1 hour incubation at RT, Alphacounts are measured.
Description: Human Bromodomain containing 4, also known as BRD4, HUNK1, or MCAP, GenBank Accession No. NM_058243, a.a. 342-460 corresponding to bromodomain 2 with N-terminal His-tag, MW = 14.7 kDa, expressed in an E. coli expression system.
Format: Aqueous buffer solution
Formulation: 50 mM Tris-HCl, pH 8.0, 138 mM NaCl, 2.7 mM KCl, 0.05% Tween-20, and 10% glycerol.
