Background: Caspase-6 belongs to the cysteine protease family of proteins, and it is considered an effector (or executioner) caspase. Caspase-6 is involved in innate immunity and inflammasome activation. It mediates pyroptosis via gasdermin D and necroptosis. Dysregulation of its activity can lead to cancer, auto-immunity and neurodegenerative disorders. This protein can cleave huntingtin (HTT) and amyloid beta-precursor protein (APP), resulting in the protein aggregates that lead to Huntington's and Alzheimer's disease, respectively. Targeting caspase-6 is thus an attractive option in disorders that are characterized by abnormal cell death pathways.
