Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
Assay Conditions: Enzyme activity was measure using Z-LYTE assay conditions (Invitrogen). Assay buffer contains 50 mM HEPES (pH 7.5), 10 mM MgCl2, 1 mM EGTA, 200 µM ATP, 0.01% Brij-35, 2 µM substrate (Tyr 04, Invitrogen), and Flt1. Reaction was performed at room temperature for 1 hour at 30°C. Developer solution B was added to the reaction and incubated for 1 hour at RT before measuring the fluorescence intensity.
