CloneID: 1.14
Antigen Long Description: This antibody was raised by immunizing female BALB/c mice with purified trout IgM.
Buffer Composition: PBS with 0.02% Proclin 300.
Chimeric Use Statement: This chimeric rabbit antibody was made using the variable domain sequences of the original mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques.
Available Custom Conjugation Options: AP, HRP, Fluorescein, APC, PE, Biotin Type A, Biotin Type B, Streptavidin, FluoroProbes 647H, Atto488, APC/Cy7, PE/Cy7
Specificity Statement: This antibody is specific for trout IgM. It reacts with the IgM heavy chain (Hedfors et al, 2012, pmid:23009920). It is not directed against nonprotein antigenic parts of the IgM, which is shown by the failure of IgM glycopeptides to inhibit precipitation (Deluca et al, 1983, pmid:6347695). 1.14 is crossreactive with atlantic salmon IgM (Hedfors et al, 2012, pmid:23009920). The antibody targets either IgM-A of IgM-B (Hedfors et al, 2012, pmid:23009920). It has been shown that this antibody does not show crossreactivity with catfish IgM (Moulana et al, 2008, pmid:18676026).
Application Notes (Clone): This antibody has been used for affinity chromatography with the protein A (Deluca et al, 1983, pmid:6347695). The antibody was shown to be effective in flow cytometry application (Deluca et al, 1983, pmid:6347695). It was demonstrated that this antibody can be utilized in the immunofluorescence staining of trout head kidney, spleen and thymus cells (Deluca et al, 1983, pmid:6347695). Furthermore, it has been shown that this antibody can be used in the immunoprecepitation of trout IgM (Deluca et al, 1983, pmid:6347695). It is used in western blots as a negative against catfish IgL σ (Edholm et al, 2010, pmid:20149930) and MHC class IIβ (Moulana et al, 2008, pmid:18676026). Finally, 1.14 antibody can be used to detect trout antibody in an ELISA (Costa et al, 2011, pmid: 21641928).
