Notes: Fluorescein-12-dUTP is a fluorescently tagged form of deoxy-UTP that can be used to generate fluorescently labeled DNA via reverse transcription, nick translation, random primed labeling, or PCR. It displays ex/em maxima of 498/517 nm, respectively. Fluorescein-12-dUTP has been used to detect nucleic acids in situ.{57903} As a starting point for labeling, utilize approximately 0.05 µg/µl DNA, 25-50 U/ml DNA polymerase, and 10-100 µM fluorescein-12-dUTP in 10 mM Tris-HCl, pH 7.5, containing 1 mM EDTA, 5 mM NaCl, and 0.1 mM DTT, as well as dATP, dCTP, dGTP, and dTTP at 1 mM each in a total volume of 20 µl.
