Applications: Study enzyme kinetics and screen small molecule inhibitors for drug discovery and high throughput screening (HTS) applications.
Background: NUAK1 (NUAK family SNF1-like kinase 1), also known as ARK5 (AMP-related protein kinase 5), is a member of the AMP-activated protein kinase family. It is found in several tissues, and it participates in cell adhesion, cell proliferation, apoptosis, and senescence. It can participate in UV-related DNA damage response by phosphorylating CDKN1A (cyclin-dependent kinase inhibitor 1 or p21). It can be activated by AKT (protein kinase B) and seems to play both cell stimulatory and inhibitory functions. For instance, it can stabilize LAST1 (large tumor suppressor kinase 1) and control senescence, but also PLK1 (Polo-like kinase 1) and stimulate S-phase progression. High expression levels of NUAK1 in ovarian cancer correlate with poor survival. It has also been shown to promote kidney fibrosis by acting as a TGF-β (transforming growth factor β)/SMAD induced activator of YAP (yes-associated protein 1). A deeper understanding of NUAK1 role and partners will help identify novel cancer therapy strategies.
Contraindications: The final concentration of DMSO in the assay should not exceed 1%.
Description: The Chemi-Verse™ NUAK1 Kinase Assay Kit is designed to measure NUAK1 (NUAK family SF-1 like kinase 1) kinase activity for screening and profiling applications using ADP-Glo™ as a detection reagent. The assay kit comes in a convenient 96-well format, with enough purified recombinant NUAK1 kinase, kinase substrate, ATP, and kinase assay buffer for 100 enzyme reactions.
Storage Stability: This assay kit will perform optimally for up to 6 months from date of receipt when the materials are stored as directed.
