Background: PDK1 (3-phosphoinositide-dependent kinase 1) is a member of the AGC serine/threonine kinase subfamily of proteins and is involved in cell survival and growth. It is involved in the activation of at least 23 downstream kinases, and it is seen as a "master kinase". It can bind to PI(3,4)P₂ (phosphatidylinositol-3,4-biphosphate) and PIP₃ (phosphatidylinositol-3,4,5-triphosphate), and translocate to the plasma membrane when PI3K (phosphoinositide 3-kinase) responds to insulin or growth-factors to produce PIP₃. Its kinase activity requires trans-autophosphorylation at Ser241. It plays a crucial role in γδ cell development and activation. Studies in PDK1 knockout mice showed thymic atrophy, reduced numbers of γδ T, B and NK cells in the spleen. Overexpression of this protein is found in many cancers, such as prostate, ovarian and breast cancer. This can result in hyperactivation of AKT (protein kinase B), which in addition to cancer is also related to immune and overgrowth disorders. Abnormal levels of PDK1 also link to the development of chemoresistance, making it an attractive target in oncology. The use of BX795, a PDK1 inhibitor, resulted in sensitizing OSCC (oral squamous cell carcinoma) to chemo/radiotherapy. Further studies on PDK1 influence on the immune system may bring new cancer therapies opportunities.

Description: The Chemi-Verse™ PDK1 Kinase Assay Kit is designed to measure human PDK1 (phosphoinositide-dependent kinase-1) serine/threonine kinase activity for screening and profiling applications using ADP-Glo™ as a detection reagent. The assay kit comes in a convenient 96-well format, with enough purified PDK1, kinase substrate, ATP, and kinase assay buffer for 100 enzyme reactions.

Storage Stability: This assay kit will perform optimally for up to 6 months from date of receipt when the materials are stored as directed.
