Applications: Study enzyme kinetics and screen small molecule inhibitors for drug discovery and high throughput screening (HTS) applications.
Background: FGR is a tissue-specific member of the Src tyrosine kinase family of proteins, and it is involved in Fcγ receptor signaling pathways in phagocytosis and mitochondrial oxidative phosphorylation. This protein is overexpressed in several cancer types, such as AML (acute myeloid leukemia), and has also been linked to Alzheimer's disease (AD), Parkinson's disease (PD), pain, diabetes, and schizophrenia. Activation of the FGR/SIRT1 (sirtuin 1)/PGC-1α (peroxisome proliferator-activator receptor γ coactivator 1α) pathway may also be involved in sepsis. Interestingly, it has been found that obesity can lead to FGR activation in macrophages, and contribute to inflammation, making the development of strategies targeting the mitochondrial ROS (reactive oxidative species)- FGR axis clinically relevant in obesity. A deeper understanding of the role of this protein in health and disease will open new therapeutic avenues for several FGR linked disorders.
Contraindications: The final concentration of DMSO in the assay should not exceed 1%.
Description: The FGR Kinase Assay Kit is designed to measure FGR kinase activity for screening and profiling applications using ADP-Glo™ as a detection reagent. The assay kit comes in a convenient 96-well format, with enough purified recombinant FGR, substrate, ATP, and kinase assay buffer for 100 enzyme reactions.
Storage Stability: This assay kit will perform optimally for up to 6 months from date of receipt when the materials are stored as directed
