Application: Screen and characterize anti-IL23 antibodies
Background: IL-23, a heterodimeric complex composed of p40 and p19, is a proinflammatory cytokine produced by activated macrophages and dendritic cells. This cytokine plays a crucial role in immune responses by promoting the production of interferon-gamma (IFN-γ) by helper T cells and supporting the proliferation of IL-17-secreting CD4+ T cells. Due to its involvement in various autoimmune diseases, including inflammatory bowel disease (IBD), colitis, psoriasis, and arthritis, significant efforts are underway to develop reagents that can antagonize the IL-23 signaling pathway.
Description: IL-23 Responsive STAT3 Luciferase Reporter HEK293 Cell Line is an engineered HEK293 cell line expressing firefly luciferase under the control of STAT3 response element, and human IL-23 receptor complex (IL12Rβ1 NM_005535.3 and IL23R NM_144701.3). With this cell line, IL-23 activity can be monitored by measuring luciferase activity.The functionality of the IL-23 Responsive STAT3 Luciferase Reporter HEK293 Cell Line was validated in dose-response assays using a recombinant human interleukin 23 (human IL-23; p19 + p40), inhibition assays using the anti-IL-23 antibodies Ustekinumab and Guselkumab, as well as a Janus Kinase inhibitor, Ruxolitinib.
