Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
Assay Conditions: The reaction mixture contained 50 mM Tris-HCl, pH 8.0, 5 mM MgCl2, 2 mM L-Fucose, 100 µM ATP, and various amount of fucokinase in a final volume of 50 µL. The reaction was incubated at 37°C for 1 hr, and the amount of ATP consumed was calculated using Kinase-Glo Plus® reagent (Promega).
Description: Human fucokinase (GenBank Accession No. NM_145059), a.a. 2-1084(end) with N-terminal FLAG-tag, MW = 118.6 kDa, expressed in baculovirus using an Sf9 insect cell expression system.
