Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. 

Assay Conditions: Add 50 µl reaction mix (20 mM phosphate buffer, pH 7.4 and LSD1) to microplate wells coated with the histone peptide substrate. Incubate at room temperature for 30 min. Add antibody against demethylated substrate, incubate 1 hr. Then, add secondary HRP-labeled antibody and incubate for 30 minutes. Finally, add HRP chemiluminescent substrates and read luminescence.

Description: Variant 2 Human LSD1 [also known as KDM1A (lysine (K)-specific demethylase 1A) or AOF2, amine oxidase (flavin containing) domain 2)] GenBank Accession No. NM_015013, a.a. 158-end, MW=77.9 kDa, expressed in an E. coli expression system with N-terminal GST-tag and PreScission Protease site. GST-tag was cleaved after affinity purification.

Format: Aqueous buffer solution

Formulation: 40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, 1 mM DTT, 1 mM EDTA, 0.05% Tween-20, and 20% glycerol.
