Encompassing Amino Acids: full length

Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. 

Assay Conditions: 5 mM MOPS, 2.5 mM β-glycerophosphate, 5 mM MgCl2, 1 mM EGTA, 0.4 mM EDTA, 0.05 mM DTT, 2 mM ATP. Incubate RSK2 with 0.2 mg/ml RSK2 peptide substrate and 50 µM [33P]-ATP at 30°C for 15 minutes, then spot reaction on phosphocellulose paper, fix in 1% phosphoric acid, and assay with a scintillation counter.

Background: RSK2 is a member of the RSK (ribosomal S6 kinase) family that are growth factor-regulated serine/threonine kinases. RSK2 has been shown to mediate growth factor signaling via RAS and MAPK leading to the induction of CREB serine-133 phosphorylation and activation of gene expression (1). Mutations in RSK2 have been shown to be responsible for Coffin-Lowry syndrome (CLS) which is a X-linked disorder characterized by severe psychomotor retardation, facial and digital dysmorphisms, and progressive skeletal deformations (2).

Description: Recombinant human RSK2 (ribosomal S6 kinase 2), full length, with N-terminal GST-tag, expressed by Sf9 insect cells via a baculovirus expression system.

Formulation: 50 mM Tris-HCl, pH 7.5, 150 mM NaCl, 0.25 mM DTT, 0.1 mM EGTA, 0.1 mM EDTA, 0.1 mM PMSF, 25% glycerol.
