Background: In immunoelectrophoresis and double radial immunodiffusion against normal human serum the precipitin line obtained shows a reaction of full identity with the purified immunogen. The product does not react with any other proteins component of human serum or plasma. In precipitating techniques as immunoelectrophoresis and single and double radial immunodiffusion (Mancini, Ouchterlony) to identify the presence of transthyretin or to determine its concentration. The presence of non-precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in non-precipitating antibody-binding techniques if proper controls are included.
Source: Transthyretin is the fastest protein in the immunoelectrophoresis pattern of human serum. It consists of four identical polypeptide chins with no carbohydrate. The molecular weight is 55,000 and the concentration in normal serum ranges from 100 to 400 μg/ml. It can bind thyroxine and retinal and its concentration is reduced in severe liver diseases. Freund’s complete adjuvant is used in the first step of the immunization procedure.
Specificity: Precipitating polyclonal Swine antiSerum to Human transthyretin (prealbumin).
Formulation: Delipidated, heat inactivated, lyophilized, stable whole serum. No preservative added. Total protein and IgG concentrations in the antiserum are comparable to those of normal pooled swine serum. No foreign proteins added.
