Application: Anti-GPRC5D-Anti-CD3 Bispecific Antibody can be used for studying GPRC5D+ cancer cell-mediated T cell activation, using either primary T cells or reporter cell lines such NFAT Luciferase Reporter Jurkat Cell Line (#60621).
Assay Conditions: This antibody was tested as followed. GPRC5D CHO cells (#78337) were incubated with NFAT Luciferase Reporter Jurkat cells (#60621) in a 96-well plate at a 1:2 ratio. Anti-GPRC5D-Anti-CD3 Bispecific Antibody and the negative control Anti-Claudin18.2-Anti-CD3 IgG Bispecific Antibody (#101541) were serially diluted and added to the cells. After 20 hours, luciferase activity was measured with ONE-Step™ Assay System (#60690) and luminescence was measured in a plate reader.
Background: The orphan G protein-coupled receptor, class C group 5 member D (GPRC5D) is a retinoic acid-inducible protein with seven transmembrane segments and a short N-terminal extracellular region. The strong and selective expression of GPRC5D in Multiple Myeloma (MM) cells, similar to the expression pattern of B-cell maturation antigen (BCMA), makes it a promising target for the treatment of MM. It is anticipated that GPRC5D will be an alternative CAR (chimeric antigen receptor)-T cell target for MM immunotherapy.
