Applications: Useful for the study of bromodomain binding assays, screening inhibitors, and selectivity profiling.
Assay Conditions: Enzyme was assayed according to BPS BRD3 (BD2) Assay Kit (Cat# 32523) protocol. BRD3 protein is incubated with biotinylated acetylhistone peptide substrate (Cat. #33000) in BRD assay buffer (Cat. #33001) in a 10 µl reaction for 1 hour at RT. AlphaLISA® (R) GSH acceptor beads (PerkinElmer) are added, followed by Alphascreen®(R) Streptavidin-conjugated donor beads (PerkinElmer). After 1 hour incubation at RT, Alphacounts are measured.
