Application Note: CDK5P25 Protein is stored in 50mM Tris-HCl, pH 7.5, 150mM NaCl, 10mM glutathione, 0.1mM EDTA, 0.25mM DTT, 0.1mM PMSF, 25% glycerol. CDK5P25 Protein is suitable for use in Western Blot. Expect a band approximately ~ 49kDa on specific lysates or tissues. Specific conditions for reactivity should be optimized by the end user.

Purity and Specificity: Recombinant human CDK5R1 (98-end) was expressed by baculovirus in Sf9 insect cells using an N-Terminal Glutathione-S-Transferase fusion protein. The purity was determined to be >85% by densitometry.

Background: CDK5P25 (cyclin-dependent kinase 5, regulatory subunit 1) is a neuron-specific activator of CDK5 and is required for proper development of the central nervous system (1). The p35 form of this protein is proteolytically cleaved by calpain, generating a p25 form of the protein which relocates from the cell periphery to nuclear and perinuclear regions. CDK5P25 regulates CDK5 activity by prolonging its activation and changing its cellular location and this may be important in pain signaling (2). The CDK5P25 accumulates in the brain neurons of patients with Alzheimer's disease and this accumulation correlates with an increase in CDK5 kinase activity which may result in aberrantly phosphorylated forms of the microtubule-associated protein tau. CDK5P25 Protein is ideal for investigators involved in Signaling Proteins, Cell Cycle Proteins, Cancer, Cell Cycle, and Ser/Thr Kinases research.
