Background: IKKβ (inhibitor of nuclear factor kappa B kinase subunit beta), also known as IKK2, is a protein subunit encoded by the IKβKβ (inhibitor of kappa light polypeptide gene enhancer in B-cells, kinase beta) gene. IKKβ activates NF-κB (nuclear factor kappa-light chain enhancer of activated B cells) signaling by phosphorylating IκBα (inhibitor of NF-κB) and leading to its degradation, allowing the release and translocation of NF-κB to the nucleus. NF-κB is a family of transcription factors that regulate gene expression in a variety of tissue types, and regulate the immune system, cell proliferation, differentiation, and apoptosis. Mutations in IKKB result in defective NF-κB signaling, which has been linked to immunological disorders and cancer. The use of inhibitors towards this protein, such as SAR113945, can become a therapeutic option in autoimmune disorders and cancer.
