Encompassing Amino Acids: 285-593

Applications: Useful for the study of enzyme kinetics and regulation, to dephosphorylate target substrates and for screening inhibitors.

Assay Conditions: 20 mM HEPES, pH 7.2, 100 mM NaCl, 2 mM EDTA, 1 mM DTT, 1 mM pNPP and 2 ng/µl MEG-2. Assay was done at RT for 15 min, and the reaction was monitored at 405 nm continuously. Specific activity was calculated based on ε pNPP= 18,000 M-1cm-1.

Background: This PTP contains an N-terminal domain that shares a significant similarity with yeast SEC14, which is a protein that has phosphatidylinositol transfer activity and is required for protein secretion through the Golgi complex in yeast. This PTP was found to be activated by polyphosphoinositide, and is thought to be involved in signaling events regulating phagocytosis.

Description: Protein tyrosine phosphatase (GenBank accession number M83738), a.a. 285-593 (catalytic domain) with N-terminal GST-tag, MW=60.3 kDa, expressed in an E. coli expression system.

Format: Aqueous buffer solution

Formulation: 25 mM Tris-HCl, pH 8.0, 67 mM NaCl, 1.4 mM KCl, 50% glycerol, 10 mM glutathione, and 3 mM DTT.

Genbank: M83738

Storage Stability: >12 months at -80°C

Tags: N-terminal GST-tag

Uniprot: P43378

Warnings: Avoid freeze/thaw cycles.
