Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. 

Assay Conditions: Reaction was performed in 50 µl reaction mix containing 50 mM Tris pH 8.0, 150 mM NaCl, 0.5 mM 4-Methylumbelliferyl butyrate, and various amounts of MGLL. Reaction was incubated at 37°C for 15 minutes and fluorescent signal was measured at ex 360 nm, em 445 nm.

Background: Monoacylglycerol lipase (MAGL) catalyzes hydrolysis of intracellular triglyceride stores, forming fatty acids and glycerol. It is a key enzyme in the metabolism of 2-arachidonoylglycerol, hence is involved in a similar pathway to DAGL. MAGL is localized in the enteric nervous system where it functions to regulate levels of endocannabinoids, and as a result, regulates intestinal motility. It is also found in the gut epithelium where it may have a role in digestion.

Description: Human monoglyceride lipase (MGLL), GenBank Accession No. NM_007283, a.a. 2-313(end), with N-terminal His-tag and N-terminal GST-tag, MW=62 kDa, expressed in Sf9 insect cells via a baculovirus expression system.

Format: Aqueous buffer solution

Formulation: 40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, 228 mM imidazole, 0.04% Tween-20, 3 mM DTT, and 20% glycerol.
