Applications: Great for screening small molecular inhibitors for drug discovery and HTS applications.
Background: Cell signaling through the PD-1 receptor upon binding the PD-L2 ligand attenuates immune responses and is exploited by both tumors and viruses.
Contraindications: DMSO >1%, strong acids or bases, ionic detergents, high salt.
Description: The PD-1:PD-L2 TR-FRET Assay Kit is designed to measure the inhibition of PD-1 binding toPD-L2 in a homogeneous 384 reaction format. Cell signaling through the PD-1 receptor uponbinding the PD-L2 ligand attenuates immune responses and is exploited by both tumors andviruses. This FRET-based assay requires no time-consuming washing steps, making itespecially suitable for high throughput screening applications. The assay procedure isstraightforward and simple; a sample containing europium-labeled (Eu) PD-1, dye-labeledacceptor, biotin-labeled PD-L2, and an inhibitor is incubated for two hours. Then, thefluorescence intensity is measured using a fluorescence reader.
Storage Stability: Stable at least 6 months from date of receipt, when stored as directed. Kit components require different storage conditions. Be sure to store each component at the proper temperature upon arrival.
Supplied As: This kit comes in a convenient 384-well format, with Europium-labeled PD-1 and biotin-labeled PD-L2.
Warnings: Avoid freeze/thaw cycles.
Biosafety Level: Not applicable (BSL-1)
