Assay Conditions: 1.88 µg of PRDX1 was incubated in 50mM HEPES (pH 7.0) containing 200 µMNADPH, 3 µM thioredoxin, and 1.5 µMthioredoxin reductase. The reactionmixture was incubated at 30°C for 5min, followed by the addition of 0.22mM H2O2. NADPH oxidation wasmonitored (fluorescence decrease) for thenext 10 min.
