Background: The TFIID complex is important for coordinating the pre-initiation complex and transcription by RNA Pol II. The complex contains the TFIID protein, TATA binding protein (TBP) and multiple TBP-associated factors (TAFs). TAFII68 was identified as a subunit of the TFIID complex, but has also been found to be associated with the Pol II multiprotein complex. TAFII68 is part of the TET family of proteins that can bind RNA and single-stranded DNA. This family also includes the human sarcoma-associated proteins EWS and TLS. The association of this family of proteins with Pol II suggests that they may bridge the processes of transcription initiation and elongation. The N-terminus domain of TAFII68 in gene fusions has been shown to have oncogenic properties, similar to the chromosomal translocation associated with the EWS gene.
