Description: TPO, a growth factor specific to certain lineages, emerges from the liver, kidney, and skeletal muscle. Its primary function involves stimulating the proliferation and maturation of megakaryocytes while fostering an increase in circulating platelet levels in vivo. TPO transmits its signals through the c-mpl receptor, playing a crucial role as a regulator of circulating platelets. Both human and murine TPO display cross-species reactivity. The Human TPO gene encodes a 353 amino acid glycoprotein featuring a 21 amino acid signal sequence, a 15 amino acid erythropoietin-like domain, and a highly glycosylated 179 amino acid C-terminal domain. Recombinant Human TPO, comprising 174 amino acids, forms a fully biologically active polypeptide weighing 18.6 kDa, incorporating the erythropoietin-like domain from the full-length TPO protein.
