Application: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. 

Assay Conditions: Assay was done in a Kinase buffer (BPS Bioscience #79334) using MBP (0.2 mg/ml) as a substrate with 20 μM ATP at 30°C for 45 minutes. The amount of ATP transferred was calculated using Kinase-Glo reagent (Promega).

Background: RIPK2 (receptor-interacting protein kinase 2) is a serine-threonine kinase that has been shown to mediate the activation of pro-inflammatory signals by NODs (Nucleotide-binding oligomerization domain-containing proteins), suggesting RIPK2 may be a promising therapeutic target in autoimmune and inflammatory diseases. This protein is expressed in several tissues, such as the spleen, placenta, testis and heart. It can interact with TNFR (tumor necrosis factor receptor) and thus participate in NOD and TLR (Toll-like receptor) pathways. Inhibition of RIPK2 may result in therapeutic benefits, and most of the inhibitors developed so far involve binding to the ATP site. Further studies and development will result in therapeutical benefit for RIKP2 related disorders.

Description: Recombinant human RIPK2 (receptor interacting kinase 2), encompassing amino acids 2-299. This construct contains an N-terminal GST tag followed by a PreScission Protease Cleavage Site. The recombinant protein was affinity purified.
