Notes: Cayman’s sPLA2 (Type V) Inhibitor Screening Assay Kit provides a robust and easy-to-use platform for identifying novel inhibitors of human sPLA2 type V. The assay uses diheptanoyl thio-PG, a sulfur-containing analog of phosphatidylglycerol, which is the preferred substrate of sPLA2 type V. Upon hydrolysis by sPLA2 type V at the sn-2 position, the free thiol can be detected by the sPLA2 Assay Probe. This results in a product that can be easily quantified by measuring absorbance at a wavelength of 410 nm. A potent and reversible inhibitor, LY315920 (included in this kit as sPLA2 Control Inhibitor), inhibits the enzyme with an IC50 value of approximately 100 nM. As an alternative, Cayman offers sPLA2 (Type V) Inhibitor Screening Kit (PC Substrate) (Item No. 10004883), which uses a PC-based substrate.
