Antigen Long Description: F11.2.32 was prepared by generating a hybridoma cell line in which myeloma cells were fused with splenocytes derived from BALB/c mice immunized with HIV-1 PR in the presence of Freund's adjuvant. F11.2.32 was selected for its ability to bind to HIV-1 PR by ELISA.

Specificity Statement: F11.2.32 Fab binds to HIV-1 PR with a dissociation constant of 5 nM (determined by SPR at pH5.2) at an epitope consisting of MSLPGRWKPKM (aa 36-46). HIV-1 PR is a homodimeric apartate protease that specifically cleaves the viral Gag and Gag/Pol polyprotein precursor. A correlation has been observed between the activity of the protease and the degree of infectivity, and so this protein is a major antiviral target.

Application Notes (Clone): The IgG and Fab variants of F11.2.32 are both able to strongly inhibit the catalytic activity of HIV-1 PR (IC50 ~35 nM). Inhibition occurs by distorting the native structure of the HIV-1 PR at the base of the flap region, which plays an important role in regulating access to the active site. This antibody is also able to detect HIV-1 PR by ELISA.
