Assay Conditions: Assay was run according to CDK12/Cyclin K Kinase Assay Kit (BPS Bioscience #78298) in a Kinase buffer containing 2 mM DTT using 0.4 mg/ml CDK12/CyclinK substrate as a substrate with 10 µM ATP at 30°C for 90 min. The amount of ATP transferred from autophosphorylation as well as substrate phosphorylation was calculated using Kinase-Glo reagent (Promega).
Description: Recombinant human Complex of CDK19 and Cyclin. Recombinant human CDK1 (cyclin dependent kinase 12), encompassing amino acids 696-1082. This construct contains an N-terminal GST-tag and includes a Thrombin Cleavage Site to facilitate removal of the GST portion. Recombinant human Cyclin K (CCNK), full length, encompassing amino acids 2-580(end). This construct contains an N-terminal GST-tag and includes a Thrombin Cleavage Site to facilitate removal of the GST portion. These two recombinant proteins were co-expressed and affinity purified.
