Background: ABL2, also known as Abelson-related gene protein or ABL proto-oncogene 2, is a cytoplasmic tyrosine kinase of the Abelson tyrosine kinase family. ABLs are involved in development and cellular homeostasis and become abnormally activated in the presence of insults, such as inflammation, tissue injury, cancer, neural degeneration and other diseases. They respond to growth factors, chemokines, oxidative stress, and DNA damage by promoting cytoskeleton rearrangements. For instance, in solid tumors their activation can lead to invadopodia formation and EMT (epithelial-mesenchymal transition), which promotes metastasis and resistance to chemotherapy. Inhibition of ALB2 by allosteric inhibitors and PROTACs may prove beneficial in creating sensitivity of the patients to standard methods of cancer treatment. A deeper understanding of the mode of action of ABL2 and results of its inhibition will move the cancer therapy field forward.
Contraindications: The final concentration of DMSO in the assay should not exceed 1%.
