Background: HCK (hematopoietic cell kinase) is a member of the Src non-receptor tyrosine kinase (NRTKs) family of proteins, mainly found in myeloid cells. It is localized in the cytoplasm of cells in a phosphorylated inactive form. Activation of cytokine receptors leads to phosphatase-induced dephosphorylation of HCK, resulting in its stimulation. It phosphorylates downstream partners, including PI3K (phosphoinositide 3-kinase)/AKT and STAT5 (signal transduced and activator of transcription 5). It is involved in inflammation, phagocytosis, membrane remodeling and cell migration. It is also linked to cancer, being part of the CXCL12 (C-X-C motif chemokine 12)/CXCR4 (C-X-C chemokine receptor type 4) signaling in AML (acute myeloid leukemia). It can bind to NLRP3 and induce caspase-1 activation in vitro. The excessive activation of the NLRP3 (NOD-, LRR- and pyrin domain containing protein 3) inflammasome is involved in diseases such as type 2 diabetes, atherosclerosis, MS (multiple sclerosis), Parkinson's, and Alzheimer's disease. The use of HCK inhibitors is promising in the treatment of inflammasome-related disorders and cancer.

Description: The Chemi-Verse™ HCK Kinase Assay Kit is designed to measure human HCK (hematopoietic cell kinase) tyrosine kinase activity for screening and profiling applications using ADP-Glo™ as a detection reagent. The assay kit comes in a convenient 96-well format, with enough purified HCK (amino acids 230-497), kinase substrate, ATP, and kinase assay buffer for 100 enzyme reactions.
