Background: CLEC2D (C-type lectin domain family 2 member D), also known as LLT1 (lectin like transcript -1) belongs to the NK cell receptor C-type lectin family of proteins. It functions as a ligand for the KLRB1 receptor (killer cell lectin-like receptor subfamily B member 1), also known as CD161, involved in sell recognition responses in NK cells. It is found in monocytes, NK cells and certain T cells. It plays a role in tumor and viral immunity and anti-inflammatory responses. It is also expressed in tumor cells, and the interaction between CLEC2D and CD161 in NK cells results in decreased cytotoxicity and IFNγ (interferon gamma) secretion. In the context of inflammation, CLEC2D transports DNA-histone complexes to endosomes and triggers TLR9 (toll like receptor 9) responses in macrophages and microglia. A deeper understanding on the roles of this protein can aid in the development of therapies for diseases that involved CLEC2D and its partners
