Background: The physiological activity for which growth hormone is best known is the promotion of growth of bone, cartilage, and soft tissues. Detectable levels of growth hormone are found throughout the remainder of adulthood, suggesting other functions in addition to promotion of growth. Growth hormone may be important for the maintenance of lean body mass, most growth-promoting effects of growth hormone are mediated by IGF-1 the synthesis of which is regulated by growth hormone. The biological activities of growth hormone are mediated by receptors belonging to one of the cytokine receptor families. Growth hormone has been shown to be produced by T- cells, B-cells, and macrophages. In human lymphocytes, growth hormone appears to up-regulate its own expression. Growth hormone appears to act as an enhancer of immune responses and is produced in considerable amounts by T- helper cells.

Biological Activity: Determined by a dose-dependent cell proliferation assay using the rat lymphoma, Nb2-11, to be in the range of 0.1 ng/ml.

Description: Recombinant human growth hormone is a disulfide-linked monomeric protein consisting of 192 a.a. and migrates as an approximately 22 kDa protein under reducing and non-reducing conditions. Optimized DNA sequence encoding human growth hormone mature chain was expressed in E. coli.

Endotoxin Level: <0.1 ng/µg (1 EU/µg), using the LAL gel clot method.

Format: lyophilized protein

Formulation: Lyophilized from a 0.2 µm filtered solution of 5 µM NaHCO3, pH 10.

Genbank: P01241

Purity: ≥95% by SDS-PAGE and HPLC

Reconstitution: Reconstitute at 0.1-1.0 mg/ml in distilled water. This solution can then be diluted into other buffers. To maximize product collection from vial surface, vortex briefly and then spin down to recollect the liquid.

Storage Stability: The lyophilized protein is stable for at least 2 years from date of receipt when stored at -20°C. Upon reconstitution, store in working aliquots at +4°C for up to one month, or at -20°C for up to six months, in the presence of a carrier protein. Avoid repeated freeze/thaw cycles.

Uniprot: P01241

Warnings: Avoid freeze/thaw cycles.

Biosafety Level: Not applicable (BSL-1)
