Assay Conditions: Assay done using Alphascreen® technology, using BPS JMJD2B Homogeneous Assay Kit, # 50414. JMJD2B enzyme (60 ng/µl) is incubated with biotinylated histone H3 peptide substrate in JMJ assay buffer in a 10 µl reaction (50 mM HEPES (pH 8.0), 1 mM α-ketoglutarate, 80 µM FeSO4, 2 mM ascorbic acid) for 1-2 hours at RT. AlphaLISA® anti-Mouse Acceptor beads (PerkinElmer) are added, followed by the primary antibody addition and 30 min incubation at RT. Alphascreen® Streptavidin-conjugated donor beads (PerkinElmer) are added. After 10-15 minute incubation at room temperature, Alphacounts are measured.
