Assay Conditions: Assay buffer: 25 mM Tris pH 9.0, 25 µM ZnCl2, 0.05% Tween-20 Enzyme was diluted to 100 ng/µl in a solution containing 1 mM AMPA (amino-phenyl mercuric acetate) in assay buffer activated for 1 hour at 37°C. Then, 25 µl of 2X 2-fold serial dilution enzyme titration in assay buffer was mixed with 25 µl of 20 µM substrate solution (Mca-RPKPVE-Nval-WRK(Dnp)-NH2 Fluorogenic MMP Substrate, R&D Systems, ES002, 10 mg/ml = 6 mM stock in DMSO, 10 µM final assay concentration) in a Corning 384-well HV black microplate. Fluorescence was kinetically recorded at 320 nm excitation/405 nm emission for 30 minutes.
