Amino Acid Sequence: FNLPL GNYKKPKLLY CSNGGHFLRI LPDGTVDGTR DRSDQHIQLQLSAESAGEVY IKGTETGQYL AMDTEGLLYG SQTPNEECLF LERLEENHYN TYTSKKHAEK NWFVGLKKNG SCKRGPRTHY GQKAILFLPL PVSSD

Background: aFGF and bFGF display 55% homology which is limited, however, to a few functional domains. The structure of aFGF, revealed by X-ray crystallography, shows a similar folding structure as IL-1. Because aFGF binds to the same receptor as bFGF, aFGF displays more or less the same spectrum of activities. It is, however, generally less active than bFGF in similar assays. FGF receptors are encoded by a gene family consisting of at least four receptor tyrosine kinases that transduce signals important in a variety of developmental and physiological processes related to cell growth and differentiation.

Biological Activity: The ED50 as determined by the dose-dependent stimulation of thymidine uptake by BaF3 cells expressing FGF receptors is ≤ 10 ng/ml, corresponding to a specific activity of≥1 x 10^5 units/mg.

Description: Recombinant murine aFGF (acidic Fibroblast Growth Factor) is a disulfide-linked monomer protein consisting of 141 a.a. and migrates as an approximately 16 kDa protein under reducing and non-reducing conditions. Native murine aFGF, generated by the proteolytic removal of the signal peptide and propeptide, has a calculated mass of 16 kDa. Optimized DNA sequence encoding murine acidic Fibroblast Growth Factor mature chain was expressed in E. coli.

Endotoxin Level: <0.1 ng/µg (1 EU/µg), using the LAL gel clot method.

Format: lyophilized protein

Formulation: Lyophilized from 0.2 µm filtered PBS solution.

Genbank: P61148

Purity: ≥95% by SDS-PAGE and HPLC

Reconstitution: Reconstitute at 0.1-1.0 mg/ml in distilled water. This solution can then be diluted into other buffers. To maximize product collection from vial surface, vortex briefly and then spin down to recollect the liquid.

Storage Stability: The lyophilized protein is stable for at least 2 years from date of receipt when stored at -20°C. Upon reconstitution, store in working aliquots at +4°C for up to one month, or at -20°C for up to six months, in the presence of a carrier protein. Avoid repeated freeze/thaw cycles.

Uniprot: P61148

Warnings: Avoid freeze/thaw cycles.

Biosafety Level: Not applicable (BSL-1)
