Background: NcoR2 (nuclear receptor co-repressor 2), also known as SMRT (silencing mediator for retinoid or thyroid-hormone receptor), is a transcriptional co-regulatory protein that can recruit histone deacetylases to promoter regions. It is expressed widely in humans and its role in the regulation of different genes makes it critical for the development and maintenance of tissues. NcoR2 has been linked to multiple cancer types, acting as an oncogene or tumor suppressor depending on context. For instance, in breast cancer, it can lead to high levels of cyclin D and cell proliferation. It has also been implicated in treatment resistance, by inhibiting cytotoxic stress response and anti-tumor immunity by regulating HDAC3 (histone deacetylase 3). Targeting NcoR2-HDAC3 using an AAV NcoR2-HDAC3 competitor resulted in better responses to chemo- and immune checkpoint therapy, opening new therapeutic windows in oncology.
