Activity: The fibroblast growth factor receptors (FGFRs) are cell surface receptors with intrinsic tyrosine kinase activity, which is necessary for receptor activation and signal propagation. PD173074 is a potent and selective inhibitor of FGFR tyrosine kinase activity, blocking autophosphorylation of FGFR1 and FGFR3. However, poorly inhibits PDGFR and c-Src and has no effect on EGFR, InsR, MEK, or PKC.
Effect: PD173074 inhibits proliferation and differentiation of oligodendrocyte progenitors. PD173074 also blocks tumor growth in H510 and H69 SCLC xenograft models.
