Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. 

Assay Conditions: A 50 µl reaction is conducted in abuffer containing 50 mM Tris (pH 7.4), 10mM MgCl2, 50 mM KCl, 2 mM ADP, 10 mMphosphoenolpyruvate (PEP) at roomtemperature for 15 min. ATP production isdetected using Kinase-Glo® LuminescentKinase Assay Platform.

Background: PKLR is a pyruvate kinase which catalyzes the transphosphorylation of phohsphoenolpyruvate into pyruvate and ATP. This reaction is the rate-limiting step of glycolysis. PKLR gene encodes the L- and R-type isoenzymes through alternate splicing events controlled by different promoters. L-type isoform can also appear as a tetramer and is upregulated by glucose with implications in maturity-onset diabetes of the young.
