Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. 

Assay Conditions: A 50 µl PKM2 reaction is conducted in abuffer containing 50 mM Tris (pH 7.4), 10mM MgCl2, 50 mM KCl, 2 mM ADP, 10mM phosphoenolpyruvate (PEP) at roomtemperature for 15 min. ATP production isdetected using Kinase-Glo® LuminescentKinase Assay Platform.

Background: PKM1 and PKM2 are different splicing products of the PKM gene (exon 9 for PKM1 and exon 10 for PKM2). The M1 isoform (PKM1) is expressed in muscle and brain, and the M2 isoform is expressed during embryogenesis, in adipose tissue and pancreatic islets, and is the predominant form found in cancer cells.

Description: Human Pyruvate kinase M2 (PKM2) also known as PKM1/M2, isoform a (GenBank Accession No. NM_002654) full length with N-terminal His-tag, MW = 58.9 kDa, expressed in an E. coli expression system.
