Applications: Useful for the study of enzyme kinetics and regulation, to dephosphorylate target substrates and for screening inhibitors.
Assay Conditions: 20 mM HEPES, pH 7.2, 100 mM NaCl, 2 mM EDTA, 1 mM DTT, 1 mM pNPP and PTPase. Assay was done at RT for 15 min, and the reaction was monitored at 405 nm continuously. Specific activity was calculated based on ε pNPP= 18,000 M-1cm-1.
Background: Protein tyrosine phosphatase nonreceptor, type 1 (PTPN1) is the founding member of the protein tyrosine phosphatase (PTP) family. PTPN1 catalyze the hydrolysis of the phosphate monoesters specifically from tyrosine residues. PTPN1 is known to be involved in signaling pathways that regulate a variety of cellular processes including cell growth and oncogenic transformation. PTPN1 also has been shown to act as a negative regulator of insulin signaling. PTPN1 has also been reported to dephosphorylate epidermal growth factor receptor kinase, which implicated the role of this phosphatase in cell growth control and cell response to interferon stimulation.
