Background: The defined antibody reactivity is restricted to beta-2 microglobulin as tested at the level of sensitivity of immuno-precipitation techniques. A single precipitin line is obtained with foetal serum, colostrum and selected pathological plasma and urine containing high levels of beta-2 microglobulin in bidimensional electrophoresis, immunoelectrophoresis and double radial immunodiffusion (Ouchterlony) which shows a reaction of full identity with the purified immunogen. In precipitating techniques as immunoelectrophoresis, single and double radial immunodiffusion (Mancini, Ouchterlony) to identify the presence of beta-2 microglobulin and to determine its concentration. The presence of non-precipitating antibodies has not been assayed. It does not exclude the use of the antiserum in non-precipitating techniques if proper controls are included.
Source: Beta-2 microglobulin is a member of the Immunoglobulin superfamily. The amino acid sequence resembles that of immunoglobulin constant domains. It is isolated from urine of patients with impaired readsorption in the kidney tubules. High levels of beta-2 microglobulin are as present in cord blood serum and foetal serum. The host animal is immunised with highly purified beta-2 microglobulin. Freund’s complete adjuvant is used in the first step of the immunization procedure.
Specificity: Precipitating polyclonal Rabbit antiSerum to Human beta-2 microglobulin.
Formulation: Delipidated, heat inactivated, lyophilized, stable whole serum. No preservative added. Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal rabbit serum. No foreign proteins added.
