CloneID: 0A07
Antigen Long Description: The original antibody was generated by immunizing alpacas with heat-inactivated Salmonella cells.
Buffer Composition: PBS with 0.02% Proclin 300.
Chimeric Use Statement: This is a chimeric antibody, developed from the original camelid VHH antibody, specifically engineered for enhanced compatibility with existing reagents, assays, and techniques.
Available Custom Conjugation Options: AP, HRP, Fluorescein, APC, PE, Biotin Type A, Biotin Type B, Streptavidin, FluoroProbes 647H, Atto488, APC/Cy7, PE/Cy7
Specificity Statement: This antibody is specific for the flagellin protein (FliC) of Salmonella enterica, which plays a pivotal role in bacterial motility and pathogenicity. FliC is the main structural component of the bacterial flagella in Salmonella enterica, tail-like structures essential for bacterial movement and invasion of host tissues. Targeting FliC can potentially neutralize the pathogen by hindering its motility, thus preventing its invasion of host cells. Additionally, antibodies against FliC may facilitate bacterial recognition and elimination by the host immune system, aiding in the clearance of the bacteria from the body.
Application Notes (Clone): The original version of this antibody (alpaca VHH) exhibited the highest inhibition of intracellular S. enterica (strain Hadar 5643) growth out of all of the tested antibodies. In vitro, the antibody was evaluated for its ability to disrupt Salmonella colonization in chicken intestinal cultures, resulting in a significant 7.5-log reduction in Salmonella genome copy numbers in ileum sections. In vivo, it was tested for its protective efficacy in broiler chickens challenged with Salmonella enterica via oral gavage. The antibody significantly decreased Salmonella levels by 0.7 log10 in both jejunum and ileum compared to the control group when administered at three different time points post-challenge (US10421806B2).
